5、 和雷帕霉素处 理组 （ 口服雷帕霉素 ） ，4 周后处死动物。HE 染色观测血管病变， 免疫组化检测 增生内膜中 SDF-1 的表达，小室迁移系统观察 SDF-1 对平滑肌细胞迁移的影 响 ，酶联免疫吸附试验检测血清 SDF-1 水平 。细胞实验：原代培养 SD 大鼠 胸主动脉血管平滑肌细胞（VSMC）， MTT 法检测不同浓度的雷帕霉素（0、1、 10 、 100ng/ml） 对 VSMC 增殖的影响， transwell 小室法检测不同浓度的雷帕霉素 （0、1、10、100ng/ml）对 VSMC 迁移的影 响，RT-PCR 和 Western blot 分别检 测不同浓度的雷帕霉素 （
6、0、1、10、100ng/ml）对 VSMC SDF-1 mRNA 和蛋白 表达的影响。 结果 ： 球囊拉伤明显诱 导血管内膜增生， 增生 内膜中有明显的 SDF-1 的表 达，雷帕霉素能显著下调增生内膜 和血清中 SDF-1 的表达，且 SDF-1 能浓度 依赖性地诱导平滑肌细胞的迁移 ；MTT 法测定结果表明 1、10、100ng/ml 的雷 帕霉素都能显著抑制 VSMC 增殖 （p 0.05 或 p 0.01）； transwell 小室法检测结果 表明 1、 10、 100ng/ml 的雷帕霉素都能显著抑制 VSMC 迁移 （p0.05 或 p0.01）， 且呈浓度依赖性；RT-PCR

8、；表达 - 5 - Effect of Rapamycin on Stromal Cell-Derived Factor-1 Alpha Expression and Migration of Rat Vascular Smooth Muscle Cell ABSTRACT OBJECTIVE To investigate the anti-restenosis effect of rapamycin and its effect on stromal cell-derived factor 1 alpha(SDF-1). METHODS The animal models of rat ca
9、rotid artery intimal hyperplasia was established with balloon injury. 20 twelve weeks old rats was randomly divided into two groups: baseline group (n = 10) and rapamycin treatment group (n = 10, 25 mg/kg Rapamycin). HE staining was used to observe vascular lesions. Immunochistochemistry staining wa
10、s used to detect the expression of SDF-1 in lesions. The migration of smooth muscle cells induced by SDF-1 was observed with transwell. The levlel of blood serum SDF-1 was measured by ELISA, Proliferation of vascular smooth cells was measured by methyl thiazolyl tetrazolium （MTT ） method, Effect of different concentration of rapamycin(0 、 1、 10、 100ng/ml) on serum induced migration of vascular smooth cells was tested by transwell, Measurement of SDF-1in mRNA and protein level was done by RT-PCR and western blot respectively.
