椒样薄荷脱毒苗的规模化生产研究 郭丹丽 王自健 蒋新明 路喆 李敏 徐盼盼 王朴摘要[目的]更新伊犁地区的椒样薄荷品种,对伊犁地区椒样薄荷进行提纯及复壮[方法]采用组培技术对椒样薄荷品种进行茎尖脱毒、离体快繁及大规模生产以椒样5号为材料,研究大规模生产下椒样薄荷茎尖分化、增殖、生根的最适激素浓度[结果]可使用白砂糖和琼脂条代替蔗糖、琼脂,降低经济成本;大规模生产时,最适椒样薄荷增殖培养基为1/2 MS+0.5 mg/L 6-BA+0.01 mg/L NAA,最适生根培养基为1/2 MS+0.10 mg/L NAA[结论]大规模组培不同阶段所使用基本培养基不同,激素组合也不同,大规模生产时以经济节约为主;利用茎尖快繁技术可快速更新换代椒样薄荷品种关键词椒样薄荷;脱毒苗;组织培养;提纯;复壮S567.23+5A0517-6611(2017)17-0122-02Abstract[Objective]To update the peppermint cultivated varieties in Yili area, purify and rejuvenate Yili peppermint. [Method]Shoot tip detoxification, in vitro rapid propagation and mass production of peppermint varieties were conducted by tissue culture technology.Using No.5 pepper as the material, the optimum hormone concentration of peppermint tip differentiation, proliferation and rooting in large scale production was studied.[Result]Sugar and agar bars can be used instead of sucrose and agar to reduce economic costs. In mass production, the optimum proliferation medium of peppermint was 1/2 MS+0.5 mg/L 6BA+0.01 mg/L NAA, and the optimum rooting medium was 1/2 MS+0.10 mg/L NAA. [Conclusion]The basic medium used in different stages of tissue culture was different, and the hormone combinations were different.
