3、investigate the chemical hypoxia effect of cobalt chloride on the cell injury,proliferation, and the expression of mRNA of soluble vascular endothelial growth factorreceptor-1(sFlt-1) of human umbilical vein endothelial cells.Methods: cell injury was evaluated by cell viability (MTT assay), lacticde

4、hydrogenase (LDH) released from all group HUVECs into the medium were measuredby using LDH kits, and the expression of sFlt-1 of all group was detected by using thequantitative reverse transcriptase-polymerase chain reaction (RT-PCR).Results: cobalt chloride could injury HUVECs, induced the expressi

5、on of mRNA ofsoluble vascular endothelial growth factor receptor-1(sFlt-1), and there was a timedependent manner.Conclusion: HUVECs were injured induced by COCl2, proliferative ability waslessened and the expression of sFlt-1 mRNA was up-regulated, the expression of sFlt-1mRNA of HUVECs may play an

8、Cl2Methods:The cells had been cultured under serum starvation condition for 12 hour,which were randomly divided into the following groups:normal control group (cells were incubated under normal condition);hypoxic group (The cells were incubated under condition of the 21 umol concentration of CoCl2fo

9、r 12hour, 24hour and36hour respectively);hypoxic and VEGF group (given the 100ng/L, 200ng/L, 300ng/L concentration ofVEGF to above group and kept the incubational time as those);VEGF control group.( given the 100ng/uL,200ng/uL,300ng/uL concentration ofVEGF to cells and incubated under normal conditi

10、ons for 12hour, 24hour and36hourrespectively);Detect the expression of sFlt-1 of all group by using the quantitative reversetranscriptase-polymerase chain reaction(RT-PCR), lactic dehydrogenase (LDH) releasedfrom all group HUVECs into the medium were measured by using LDH kits,cell injurywas evaluat

The expression of sFlt-1 mRNA was lessened and had a concentration-dependentmanner, the mount of LDH and MTT were decreased in concomitant with the differentconcentration of VEGF, its tendency was time-and concentrati

13、on-dependent manner.3.sFlt-1 mRNA expression levels, the mount of LDH and the number of death cells hadcorrelations with each other respectively.Conclusion:1. HUVECs induced by CoCl2 had these manifestations: the release of LDH and thenumber of death cell elevated, the expression of sFlt-1 mRNA was
