3、latter is rarely used today. Epi-illuminators usually consist of a mercury or xenon lamphouse coupled to a vertical illuminator that is positioned above the main frame in a separate assembly. The microscope nosepiece and transmitted light components (diascopic illuminator, condenser, field diaphragm
4、, filters, etc.) are built into the main frame, while fluorescence components are housed in the vertical illuminator. These illuminators often contain a revolving or sliding turret that houses four to six cubes that contain a mixture of interference filters including a barrier filter, dichroic mirro

Secondary fluorescence, emitted by fluorophores residing in the specimen, travels back through the objective and the dichroic mirror before passing through a barrier f

11、）,叶片,18,稻谷课件,注意事项,激发光挡片的使用-减少淬灭,19,稻谷课件,注意事项,荧光的淬灭与激发光的照射时间成正比，因此应尽量减少观察时间外激发光对样品的照射。激发光挡片可以部分或完全阻断激发光光路。在正式观察样品之前，激发光光路应该处于完全阻断状态。否则，在我们将样品放到载物台上，移动样品台将样品调整到物镜位置，找到要观察的细胞并进行调焦的过程中，样品上的荧光可能已经发生了相当程度的淬灭。为了避免观察前不必要的淬灭，上述一系列操作需要在透射光源下进行。 当眼睛离开物镜进行记录或将显微镜交给合作实验者观察时，要随手阻断激发光光路。实践结果表明，熟练、规范和合理的观察操作可以将样品淬灭
