1、血吸虫感染小鼠及家兔抗Sj23kDa膜蛋白大亲水肽段(Sj23HD)的抗体IgG应答模式及其免疫诊断价值张伟 ,王玠 ,余传信 *,宋丽君,殷旭仁,钱春艳,沈双,金一,柯雪丹,姚媛,高玒,许永良,杨静寄生虫病预防与控制新技术研发室,卫生部寄生虫病预防与控制技术重点实验室,江苏省血吸虫病防治研究所,无锡,214064,中国摘要 目的 探讨日本血吸虫中国大陆株 23kDa 膜蛋白分子大亲水肽段(Sj23HD)的抗体反应模式及重组 Sj23HD-HSA 用于血吸虫诊断和疗效考核的价值。方法 构建血吸虫尾蚴感染小鼠及家兔模式动物,采集尾蚴感染前,感染后、治愈后各不同时间点小鼠及家兔血清。以酵母表达的纯

2、重组 Sj23HD 蛋白为抗原,通过间接 ELISA 方法,检测感染前、感染后与治愈后不同时间点血吸虫感染小鼠和家兔血清中抗 Sj23HD 抗体 IgG 应答水平的动态变化,观察 Sj23HD在小鼠及家兔的抗体反应应答模式及其疗效考核潜能。同时,通过间接 ELISA方法,以 Sj23HD-HSA 融合蛋白分别检测 132 份血吸虫、 30 份肝吸虫、10 份肺吸虫、以及 80 份健康人血清样本,观察该重组抗原用于血吸虫病诊断的价值,并将抗可溶性虫卵抗原的抗体应答反应作为对照。结果 血吸虫感染小鼠及家兔体内抗 Sj23HD 蛋白的抗体反应呈短寿抗体反应应答模式,抗 Sj23HD 抗体 IgG反应

3、随着感染时间的延长呈上升趋势,治愈后的初期继续上升,至治疗后 2-4w达到最高水平,随后逐渐下降,至治疗后 16 周,部分小鼠、家兔的抗体水平可达到阴性阈值,但下降幅度存在个体差异。抗 SEA 抗体水平在治疗后有轻度下降的趋势,而未治疗小鼠、家兔血清中抗 Sj23HD、SEA 的抗体反应均无明显下降。重组 Sj23HD -HSA 和 SEA 检测 132基金项目 国家自然科学基金(30972581) ;江苏省自然科学基金(BK2008110);国家重大科技专项(2008ZX10004-011) ;江苏省卫生厅科技项目(H201066) ;江苏省血地寄项目(X201115);江苏省预防医学项目(

Methods The sera of mice and rabbits infected with Schistosoma japonicum before infection, during infection and after cure with PZQ at different time were prepared. To

9、 observe the antibody response pattern of Sj23HD in mice and rabbits infected with Schistosoma japonicum and the potential of evaluating chemotherapeutic effect of schistosomiasis, the dynamics of antibody response against Sj23HD before infection, during infection and after cure with PZQ at differen

10、t time were detected by indirection Enzyme Linked Immunosorbent Assay (ELISA). Meanwhile, to evaluate the immunodiagnostic value of recombinant Sj23HD-HSA, sera of 132 schistosomiasis, 30 clonorchiasis, 10 paragonimiasis, 80 healthy were detected by indirection ELISA with recombinant Sj23HD-HSA. For

11、 experimental control, the antibody IgG against soluble egg antigen (SEA) in sera was detected by the same indirection ELISA in the same time. Results The antibody response against Sj23HD rose along with the infection duration extension, and enhanced continually to peak in 2-4w post PZQ treatment, t

12、hen declined gradually, the level of antibody response of partial mice or rabbits reached the negative cut off value at 16 w PZQ post treatment, but the decline extent of antibody response in different individuals was different. There was a slight decline of antibody response against SEA in mice and

13、 rabbits of treated group, but no decline of antibody response against Sj23HD and SEA in mice and rabbits of untreated group. The positive rate of antibody IgG against Sj23HD-HSA or SEA in 132 schistosomiasis sera was 88.64 percent or 97.73 percent respectively; The cross reaction rate of antibody I

14、gG against Sj23HD-HSA or SEA in 10 paragonimiasis sera was 50 percent or 90 percent respectively and in 30 clonorchiasis sera was naught percent or 16 percent respectively; No false positive of antibody IgG against both Sj23HD-HSA and SEA in 80 healthy sera was found. Conclusion The antibody respons

15、e pattern of Sj23HD protein in mice and rabbits infected with Schistosoma japonicum is shorted lived antibody response. It is potent by detecting the antibody involved in this short lived antibody response against Sj23HD-HSA for diagnosis and evaluating chemotherapeutic effect of schistosomiasis jap

16、onica.
