2、金。对取自江西某铀矿堆场的嗜酸性微生物群落优势菌菌进行了分离和初步的研究分析与探讨。本文对采自江西某矿堆浸矿堆实验的六个样品:S1,S2,S3,S4,S5,S6,在以9K+S+Fe液体培养基对其混合菌群氧化铁的效率进行分析,结果发现S3,氧化铁的效率最佳。运用双层固体平板培养技术,,以琼脂糖为凝固剂,,以SJH为底层添加菌,,分离S3样品中的微生物,,得到86种嗜酸性氧化亚铁硫杆菌(暂定名为:A1,A2,A3,A4,A5,A6),1株氧化硫硫杆菌(暂定名为:B1),3株异养菌(暂定名为:C1,C2,C3)。微生物,根据不同各菌株在不同双层固体平板表面的菌落形态特征,, 显微结构镜下观察结果,并

3、结合16SrRNA基因扩增技术对所获得的菌种进行了初步鉴定菌种。群落中占优势的嗜酸性微生物为氧化亚铁硫杆菌(Acidithiobacillus ferrooxidans,,简称A. f) 。对其中一株氧化亚铁铁硫杆菌(A6) 的培养温度、接种培养基初始PHppH值、及接种量对其生长状况的影响进行做了单因素探讨实验,。结果表明,A6氧化Fe2+最快的最适 培养温度为35,、最适初始pH值为2.3,。最合理佳的接种量为10% 。对初步研究了不同底物S、Na2S2O3和Na2SO3对其中一株氧化硫硫杆菌(B1)的不同底物对起其生长的影响做了初步研究,结果表明最适合B1的底物为Na2S2O3,其次为单
