    不同来源百合药材的质量评价及分析    刘湘丹 陈勋 刘畅宇 裴刚 周日宝 王智 陈乃宏〔Summary〕 目的 基于水分、浸出物、多糖、Regaloside A含量进行不同来源百合药材质量评价及分析,为多基原百合药材鉴别及质量评价提供依据方法 按照2015版《中华人民共和国药典》方法测定水分、浸出物;采用紫外分光光度法测定百合多糖含量;采用高效液相色谱法测定Regaloside A含量:Supersil ODS-B(250 mm×4.6 mm, 5 μm)色谱柱,柱温为30 ℃,进样量为10 μL,流速为1.0 mL/min,流动相0.1%磷酸水溶液∶乙腈=85∶15(V/V),检测波长为310 nm结果 54批百合样品水分均≤13%;43批百合样品浸出物≥18%,11批百合样品浸出物<18%;54批百合样品多糖含量区间为9.89%~23.42%;54批百合樣品Regaloside A含量区间为0.019%~0.599%结论 所有百合样品水分均符合药典要求;11批百合药材浸出物未达到药典要求,且均为龙牙百合药材;17批百合多糖含量未达湖南省地方标准;卷丹百合浸出物和Regaloside A含量均普遍高于市购龙牙百合药材。

Institute of Materia Medica, Chinese Academy of Medical Sciences and Peking Union Medical College, Beijing 100050, China)〔Abstract〕 Objective To evaluate and analyze the quality of Lilii Bulbus from different sources by the contents of moisture, bulbus extract, polysaccharides and Regaloside A, so as to provide the basis for identification and quality control of polybasic lilium. Methods Moisture and bulbus extract were determined according to the 2015 edition of Chinese Pharmacopoeia. Polysaccharides was determined by ultraviolet spectrophotometry. Regaloside A was determined by high performance liquid chromatography. The column was a Supersil ODS-B column (250 mm×4.60 mm, 5 μm). The column emperature was 30 ℃, and the injection volume was 10 μL. The flow rate was 1.0 mL/min, the mobile phase 0.1% phosphoric acid aqueous solution-acetonitrile volume ratio of 85∶15 (V/V). The detection wavelength was at 310 nm. Results The moisture contents of 54 batches of Lilii Bulbus were all less than 13%. The bulbus extracts of 43 batches of Lilii Bulbus were greater than 18%, and the bulbus extracts of the other 11 batches of Lilii Bulbus were less than 18%. The polysaccharidse contents of 54 batches of Lilii Bulbus range from 9.89% to 23.42%. The Regaloside A content range of 54 batches of Lilii Bulbus was 0.019%~0.599%. Conclusion The moisture of 54 batches of Lilii Bulbus all meets the requirements of Chinese Pharmacopoeia. The bulbus extract of 11 batches of Lilii Bulbus were unqualified, which are all from LongYa Lilii Bulbus. The mean polysaccharide content of Lilii Bulbus from different sources was not significantly different. The bulbus extract and Regaloside A contents of Juan dan Lilii Bulbus were generally higher than those of Long Ya Lilii Bulbus on the market, which provided evidence for the fact that Juan Dan Lilii Bulbus was the main medicinal material in the market. This study can provide more references for identification and quality control of Lilii Bulbus from different sources.〔Keywords〕 Lilii Bulbus; moisture; bulbus extract; polysaccharides; Regaloside A; content determination; quality evaluation百合始载于《神农本草经》,其应用历史悠久,属药食同源品种。

2015版《中华人民共和国药典》(简称《中国药典》)记载百合来源于百合科植物卷丹Lilium lancifolium Thunb.、百合Lilium brownii F.E. Brown var. viridulum Baker或细叶百合Lilium pumilum DC.的干燥肉质鳞叶[1]其味甘性寒,归心、肺经,有养阴润肺,宁心安神的功效,主要用于治疗肺虚久咳,劳嗽咳血,心烦失眠,情绪不能自主等病[1]研究表明,药用百合鳞叶中主要含有多糖、酚酸甘油酯、甾体皂苷、甾醇、黄酮、苯丙素、多糖类、生物碱及磷脂、无机盐等成分其中百合多糖含量高,具抗肿瘤、降血脂、抗病毒、抗突变和提高细胞免疫活性等多种生理功能[2-5],为百合主要功能成分之一;酚酸甘油酯亦为百合大类成分,具抗氧化、抗肿瘤、抗增殖、免疫调节及雌激素活性等生理功能[6-8],其中Regaloside A为百合酚酸甘油酯类成分之一[9],项目组前期预实验发现其在百合药材中相对百分含量较高,且具有显著的抗抑郁药理活性,具有潜在研究价值笔者收集百合药材样品过程中发现,药用百合品种主要为卷丹Lilium lancifolium Thunb.和百合Lilium brownii F.E. Brown var. viridulum Baker两种,其中卷丹以药用为主(其干燥肉质鳞叶商品习称卷丹百合),百合可药食两用(其干燥肉质鳞叶商品习称龙牙百合),湖南为以上两品种主产区,主为栽培,秋季采收;细叶百合Lilium pumilum DC.的干燥肉质鳞叶市场基本不见流通。

市场调查还发现,食用百合主流品种除龙牙百合外还有兰州百合Lilium davidi var. unicdor cotton干燥肉质鳞叶(商品习称兰州百合),其次市场还存非药用品种如金百合(其饮片性状与卷丹百合相似)入药现象,百合为多基原药材,市场存在品种难以区分或混用现象基于此,本研究在2015版《中国药典》百合药材性状鉴别及水溶性浸出物含量基础上,增加百合大类成分多糖、Regaloside A两个指标,对收集的药用百合品种、食用主流品种、药用混淆品及野生百合样品的质量进行评价和分析,以期为鉴定不同来源百合、行百合药材质量评价提供研究基础1 仪器与材料1.1  主要儀器超声波清洗机(宁波新芝生物科技股份有限公司);Waters 1525 高效液相色谱仪(配有2489紫外/可见检测器,二元泵)(美国沃特世公司);ME204E/02电子分析天平(梅特勒-托利多仪器(上海)有限公司);中草药粉碎机(天津市泰式特仪器有限公司),UV-1750紫外可见分光光度计(日本岛津);TDZ-5-A离心机(上海安亭科学仪器厂);UV1800PC紫外分光光度计(上海奥析科学仪器有限公司)1.2  主要材料无水葡萄糖(国药集团化学试剂有限公司,批号63005518);Regaloside A对照品(实验室自制,纯度≥98%);甲醇(天津市科密欧化学试剂有限公司,分析纯);磷酸(长沙分路口塑料化工厂,分析纯);乙腈(德国默克股份有限公司,色谱纯);硫酸(湖南汇虹试剂有限公式,分析纯);乙醇(安徽安特食品股份有限公司,分析纯);蒽酮(国药集团化学试剂有限公司,分析纯)。

项目组于2017-2018年度共收集54批百合样品(包括药用百合、食用百合、百合药材混伪品、野生百合),经湖南省药品检验研究院方石林教授、湖南中医药大学第一附属医院张志国教授、湖南中医药大学周日宝教授、刘塔斯教授、湖南农业大学谢红旗教授鉴定为百合科相关品种,具体信息见表1其中野生百合,为笔者野外采集,于实验室自加工用于对照2 方法与结果2.1  水分按照2015版《中国药典》第四部通则0832第二法(烘干法)对54批百合样品水分进行测定分析,54批百合样品水分含量均在13%及以下,符合药典规定2.2  浸出物按照2015版《中华人民共和国药典》第四部通则2201水溶性浸出物冷浸法,对54批百合样品进行浸出物测定,以干燥。
