EA treatment could sig2 nificantly reverse the immunosuppression and regulate the above index changes in plasma. Further

2、study showed that intracerebroventricular(icv) injection of naloxone could antagonize the depressed NK cell activity induced by trauma , and EA could also improve immunosuppression , sug2 gesting that the central endogenous opioid peptides might take part in the immunosuppression and the modulation

3、of EA. Orphanin FQ is a new member in opioid peptide family , and its role in immunosup2 pression and modulation of EA is worthy of investigation. Orphanin FQ icv attenuated immune re2 sponse envoked by trauma , and when blocking the synthesis of OFQ receptor with the antisense oligo2 nucleotide(ASO

4、) , the effect of OFQ was completely abolished , suggesting that the central orphanin FQ might participate in the modulation of the depressed immune function. However , ASO icv did not significantly change the effect of EA . The study demonstrated that central OFQ and its receptor could be downregul

5、ated or upregulated by trauma or EA , so the mechanisms of the modulation of OFQ or EA on immunosuppression induced by surgical trauma should be deeply studied. Central IL21plays an important role in neuroimmune regulation. The expression of central IL21 mRNA obviously increased after trauma. Intrac

6、erebroventricular injection of IL21antibody could im2 prove the severe immune depression induced by trauma. OFQ icv and EA could antagonize the in2 creased IL21mRNA expression in trauma rats. OFQ immuno2reactive cells and IL21mRNA were found to coexist in the cerebral cortex , hippocampus and hypoth

8、针对大鼠实验性关节炎痛阈和皮肤c2fos表达的作用及侧脑室注射孤啡肽 (OFQ)对电针效应的影响。 方法:将福氏完全佐剂(FCA)注入SD大鼠踝关节腔造成类风湿性关 节炎的模型。44只SD大鼠随机分为A模型+侧脑室注射生理盐水(NS) +电针,n= 8 ,B(模型 +侧脑室注射OFQ +电针,n= 8) ,C(正常对照,n= 8) ,D (模型,n= 10)和E (模型+电针,n=10)组。大鼠麻醉(戊巴比妥钠30 mg/ kg ,i. p. )后,参照Noble方法定位,将OFQ (20L ,50g/mL)或NS(20L)注入侧脑室(30 min内注完,保留1 min)。右侧 “太溪” 和

9、“足三里” 针刺后连接 WQ210C多用途电子穴位测定治疗仪,用疏密波形,频率20/ 100 Hz交替,电压24 V ,波宽0. 2 0. 6 ms ,持续20 min ,电针每天1次,连续5天。痛阈用气压弹簧棒测定右后足掌心皮肤加压时动 物的缩腿反应,每天1次,共测5天。取大鼠右后足垫部皮肤及皮下组织,采用免疫组织化学ABC 法测定RA模型建立后各组c2fos表达。结果与结论:电针对实验性RA关节痛具有明显的镇022针刺研究 第26卷 2001年 第3期痛效应;电针明显提高实验性RA大鼠痛阈,具有明显的后效应,注射OFQ对抗电针提高实验 性RA大鼠痛阈,而且对抗电针的后效应;电针抑制实验性R

Expression of c2fos protein in the rat plantar region was detected with immunohistochemical method. OFQ 20L , 50g/ mL was in

Results :In RA rats , the pain threshold in A , B , D and E groups was lowered significantly , while after EA treatment , the pain threshold values , particularly on the 3rd , 4th and 5th day , were increased significantly( P0. 05) , suggesting an

16、antagonizing effect of OFQ on EA analgesia.In comparison with normal control group , the positive neurons of c2fos expression in A (82. 5424. 64) and B (174. 8738. 23) , D (168. 4234. 75) and E (99. 6525. 36) groups were increased considerably in comparison with group C (0. 000. 00) , showing a striking increase of c2fos expression.
